Detection of Modified Nucleosides in tRNA from Colon Cancer Spheroids

Abstract:
Colon cancer remains one of the deadliest cancers and new molecular methods to diagnose it are needed. Colon cancer patients have differences in their levels of modified nucleic acid bases in their urine as compared to healthy patients. Transfer RNAs (tRNAs) are the most heavily modified RNA. These modifications are catalyzed by various tRNA modification enzymes. The modifications present in tRNAs have been suggested to differ from tissue to tissue within an organism, but these differences have not been well characterized. We aim to develop a mass spectrometric method which will allow for the detection of modified bases present in the tRNA of HCT 116 spheroids, a cell culture model for the colon cancer tumor.
